TC-3000G
The world's smallest gradient cycler, the TC-3000G will hold half a 96-well plate, in a horizontal format so that it offers 8 columns for annealing temperature optimisation and 6 rows for optimising reagents such as MgCl2 and primer concentrations. Annealing temperatures can be optimised over a 15°C gradient between the temperatures of 20 and 80°C.

The TC-5000 gradient thermal cycler enables you to optimise your experiments at the `touch of a screen', with the same mechanical features as the TC-4000.

TC-Plus Thermal Cyclers
The TC-PLUS thermal cyclers have all the latest features from a colour touchscreen to fast ramp rates along with some innovative additions such as an automatic, non-motorised heated lid, TERS®, thermal energy recovery system and a unique space-saving, stackable design. Progene has numerous applications in Molecular Biology and Diagnostic laboratories and is suitable for use with diagnostic kits.
Progene has been designed for the individual; priced to be inexpensive enough to be treated as an ordinary piece of laboratory equipment that everybody can own and designed to process a small number of parallel samples rapidly in unprecedented cycle times, allowing a typical 25 cycle program to be completed in around an hour, enabling the operator to achieve more PCR's in a working day.
Using advanced Peltier technology gathered from Techne's larger Cyclogene thermal cycler and developed to give the amazing heating/cooling rates of better than 2.2 - 2.8°
C/second (average cooling and heating) and using a high tolerance PRT sensor to give a temperature stability of ±
0.1°
C and a block uniformity of ±
0.3°
C.
Progene is available with heated lid as standard and a specification only sometimes found in units costing two or three times the Progene price.
One of Progene's many features is incremental/decremental segment hold time. This allows the user to increase or decrease the hold time of a given segment hold with each cycle. Use incrementally on 'extension' for techniques such as Long PCR and RAPD to allow sufficient time for the increasing quantity of DNA to extend. Use decrementally on 'denaturation' to allow initial highly specific denaturation and then less time with each cycle at 95°
C so as to maintain the enzyme's (taq) half-life. Additionally, another feature of Progene is incremental/decremental temperature used for protocols such as Touchdown PCR to optimise annealing. The interchangeable blocks available (to suit 0.5ml microcentrifuge tubes, 0.2ml micro tube/5x5 multiwell plate) can be changed in seconds.
Progene is small enough and inexpensive enough to be personal but with its technically innovative, superior robust specification, reliability and performance capabilities, suitable for the most demanding molecular scientist.

The Techne software package GENSOFT is available as an accessory for connecting the Genius to computer via the RS232 port. The software incorporates some additional features making the unit even more flexible for those who have larger throughput of samples normally requiring more than one machine or where a unit is required to interface with a robotics system.

The new Genius Thermal Cycler has been designed for the laboratory that has a high throughput requirement of samples. A wide selection of block options up to and including a 384 well microplate makes Genius the number one choice for large scale screening and genome investigations.
Regardless of the block type being used all samples are heated to a programmed temperature and held to very precise limits.

Block temperatures
To provide thermal cyclers with the high performance and specificity required for the protocols of today, heated block design and program versatility are important features.
Incremental/decremental temperatures
Touchdown PCR protocols are where a higher temperature is required for the initial cycles than later cycles. This ensures that only highly specific product is created (amplified) during the critical initial steps. A decreased, less stringent temperature is then used by gradually decreasing the annealing temperature. The effect is a higher yield and more specific product. Therefore, to eliminate the need for programming lengthy error prone protocols, choose a thermal cycler which can automatically incorporate an incremental decrease/increase in temperature.
Incremental/decremental time
Long template PCR is the amplification of large fragments of DNA (e.g. 40kb lambda, 27kb Genomic). To ensure that the extension is fully complete after each cycle, the extension time is increased with each cycle to compensate for the increased product quantity and decreasing concentrations of reagents. As above with temperature, choose a thermal cycler which can automatically increase/decrease hold time at a given temperature.
Peltier technology
The peltier effect is used in many applications for the reliable cooling of sensitive high-temperature components without the need for refrigerants or compressors. Otherwise known as thermoelectric coolers, peltiers are a special type of semiconductor that function as a solid state heat pump. By applying a low voltage DC power source to one side, heat will be moved in the direction of the current (+ to - ). The heat is thus pumped from one side of the module to the other, so that one face will be cold while the opposite face will be heated. As this effect is reversible, peltier technology is the chosen method for heating and cooling the block in the best thermal cyclers on the market today.

Adjustable speed rocking platform

Application notes:
Hybridisation protocol tips
Hybridisation techniques which involve the detection of membrane bound molecules are commonplace in most laboratories and include Northern (RNA), Southern (DNA) and Western (Protein) blot. These may come in the form of molecules transferred from a gel, slot or dot blots (where they are spatially placed on top of the membrane using a pipette or specialised manifold), or by colony/plaque adhesion. During the hybridisation process, while larger volumes of wash buffers are used, small volumes of probe and detection solutions are desired, all requiring a variety of temperatures. Below are some useful tips which improve the quality of your results.
Hybridisation vessels
To optimise your results use specially designed hybridisation incubators with rotating bottles. These offer many advantages including: reduced reagent volume, even membrane hybridisation, multiple membranes/vessel, easy probe recovery, temperature accuracy and reproducibility, safe liquid transfer.
Multiple membranes
When a large number of blots are to be hybridised with the same probe, time and reagents can be saved by placing them together. To do this, pre-warm the first reagent and place it in the pre-warmed incubation vessel before adding the membranes one by one, always ensuring that each one is fully coated before adding the next. To optimise probe: target accessibility use separation membranes between each blot.
Improving signal: noise ratios
One of the most difficult aspects of membrane hybridisation is to keep a clean particulate free blot with a low background, while still retaining a good signal:
- Temperature: Chose the correct temperature. The easiest way to decrease the background during RNA and DNA blot hybridisation procedures is to increase the temperature at which the probe and initial post-hybridisation steps are carried out. As a guide, for a longer DNA probe (e.g. 200bp) start with 65°C and for an oligonucleotide probe try 50°C. For protein detection, some primary antibodies are more specific when hybridised overnight at 4°C rather than a few hours at room temperature.
- Probes & antibodies: Take care to keep stocks of probes and detection reagents at the recommended temperatures. For non-radioactive techniques, re-using probe and antibody hybridisation solutions can lead to significant improvements of signal (store antibodies at 4°C and DNA at -20°C). When using antibodies, give the stock tubes a quick spin in a microcentrifuge and pipette from the top of the liquid to remove background causing precipitants.
- Clean equipment: Cleanliness plays an important part in membrane hybridisations. Always wear gloves, handle blots with forceps and avoid dusty environments. For non-radioactive detection techniques, microwave the reaction vessels with some water in prior to re-use as this acts to remove residues which can cause serious background problems.
- Buffers: Only use high quality reagents and ultra-pure water. When making up stocks of large volume buffers such as SSC, pass them through 0.45 µm filters to remove particulates. If using formamide, make sure it has been deionized first.
- Crosslinking: Using a UV-Crosslinker to irreversibly bind your nucleic acids to the membrane is faster and much more efficient than traditional baking and results in a higher signal.

As all users are aware, the most important temperature to be accurate is that of the primer: template annealing. As a result, using the most up-to-date technology we are able to bring you temperature uniformity across the TC-312 block within ±0.1ºC*.
As Techne specialises in temperature calibration equipment, we are also able to make available to the user the temperature accuracy of the block. This value gives the average temperature spread which may occur around the absolute "real" temperature the block is set to. The TC-312 achieves an amazing accuracy within ±0.2ºC The specifications at key temperatures can be seen in the Block Temperature Specification table.
*Uniformity is the typical variation in temperature which may occur across the block under ambient conditions of 20ºC.
